摘要:为研究环鸟甘酸-腺昔酸合成酶入核的机制,利用兔外周血B 细胞筛选抗人cGAS 蛋白单克隆抗 体.针对已三次免疫人cGAS (161-522)蛋白的新西兰兔,运用流式细胞术得到阳性效应B 细胞,在单管 里对单细胞裂解,得到总RNA,逆转录生成cDNA,两步巢式PCR 扩增同一个B 细胞来源的抗体重链和轻 链可变区序列,抗体基因扩增成功率高达80%以上.293T 重组表达兔抗人cGAS 蛋白单克隆抗体并鉴定,获 得1个具有较好结合特异性的兔抗人cGAS 蛋白单克隆抗体,ELISA 检测结果显示抗体飽效价达I : 10 00().

College of Life Sciences , Fujian Normal University , Ficzfiou 350117, China )Abstract : To investigate the mechanism how cyclic GMP-AMP synthase ( cGAS ) protein en tries into the nucleus , monoclonal antibodies against human cGAS protein from rabbit peripheral blood B cells were screened. The New Zealand rabbits have been immunized three times with human cGAS ( 161-522) fragment. Positive rabbit peripheral blood B cells were sorted by fluorescence-ac tivated cell sorting ( FACS). Single cell was lysis into individual PCR tubes , and its total RNA was obtained. With the cDNA as the template , the antibody paired heavy and light chain variable chtiins were amplified by two-step nested PCRs , the success rate of antibody gene amplification was greater than 80%. The antibodies expressed in 293T cells
were identified. We obtained a monoclonal antibody with better binding specificity. The ELISA assay showed the titer of the antibody was over 1:10 000.

兔单克隆抗体(mAb )在药物和诊断方面都具有高亲和力和特异性的优点.兔单克隆抗体相较于 鼠抗和兔多抗而言,具有更广泛的抗体谱、免疫球蛋白结构简单、更高特异性和亲和力、易于人源化 等优点.目前,有多种方法可筛选兔单克隆抗体,例如交瘤细胞技术Z :;噬菌体展示技术*5 ; B 细胞分选后基于PCR 技术筛选获得兔单抗2".然而,这些技术也有很多不足,如花费时间较长、 低效率等.近年来,随着单细胞技术的发展,兔单抗筛选技术也呈现新的发展,该技术能将单个B 细 胞的克隆扩增后,通过PCR 技术得到天然抗体的重链和轻链卜“.该技术最重要的优点就是筛选时 间短,保留重链和轻链的自然同源配对,并让抗体有产生特异性、亲和力成熟的过程.然而,单个B 收稿日期:2019-03-06 基金项目:福建省自然科学基金资助项目(2017J01621);福建省中青年教师教育科研项目(JA15130)通信作者:林清强(1971-),男,高级实验师,研究方向为生化与分子生物学.qingqianglin@ fjnu. edu. cn
