天津300070)摘要目的:建立红景天注射液中5-羟甲基糠醛高效液相谱(HPLC )含量检测方法,进一步分析多批次红景天注射液中5-羟甲基糠醛的含量,
评价其安全性。方法:采用Agilent XDB-C 18(250×4.6mm ,5μm )谱柱,0.01%磷酸水溶液为流动
相,流速为1.0mL/min ;检测波长为284nm ;柱温为30℃。结果:建立了HPLC 检测红景天注射液中5-羟甲基糠醛含量的方法,经方法学考察,符合要求。进一步分析表明,34批次红景天注射液中5-羟甲基糠醛含量范围为1.321~6.927μg/mL ,均低于药典规定的葡萄糖注射液中的5-羟甲基糠醛限量标准。结论:多数批次的红景天注射液中均含有微量的5-羟甲基糠醛,但根据临床用量比较,远小于葡萄糖注射液中5-羟甲基糠醛的限量标准,其安全性具有足够的保障。关键词红景天注射液;高效液相谱;5-羟甲基糠醛中图分类号R927.2

Objective:To establish a HPLC method for the determination of 5-hydroxymethylfurfural in Hongjingtian injection,further
analyze the content of 5-hydroxymethylfurfural in multiple batches of Hongjingtian injection and evaluate its safety.Methods:The HPLC analysis was performed on Agilent XDB C 18(4.6mm ×250mm ,
5μm)and the mobile phase consisted of 0.01%phosphoric acid aqueous solution at a flow rate of 1.0mL ·
min -1.The detective wavelength was set at 284nm and the column emperature was maiutain at 30℃.Results:The HPLC method for content determination of 5-hydroxymethylfurfural in Hongjingtian injection was established,which was tested by method and met the requirements .Further analysis shows that the content of 5-hydroxymethylfurfural in 34batches of Hongjingtian injection was in the range of 1.321~6.927μg/mL,which was lower than the standard of 5-hydroxymethylfurfural in glucose injection prescribed by pharmacopoeia.Conclusion:Most of the batches of Hongjingtian injection contain a small amount of 5-hydroxymethylfurfural,but according to the clinical dosage,it is much smaller than the limit of 5-hydroxymethylfurfural in glucose injection,and its safety is sufficient.Key words

红景天注射液可以活血化瘀,临床上主要用于
冠心病稳定型劳累性心绞痛引起的胸部刺痛、绞痛、胸闷、心悸不宁、脉细涩等[1-2]。在中药注射液的质量控制研究中,一方面有效成分的分析,如谱指纹图谱等,应用越来越普遍;另一方面,注射液中潜在的有毒物质的分析与控制,也越来越得到了重视。现有研究普遍认为,5-羟甲基糠醛是葡萄糖、果糖等单糖在高温、酸性条件或微生物的作用下发生麦拉德(Maillard )反应脱水生成的一个醛类化合
物[3-4]。随着反应的进行,食品和中药材的泽会逐
渐变深[5]。5-羟甲基糠醛对人体横纹肌及内脏有损害,具有神经毒性,能与人体蛋白质结合产生积蓄中毒[6-8]。在中国药典(2015年版)二部[9]、日本药局方(14版)、欧洲药典(7.0版)中葡萄糖注射液项下中均用UV 法来控制其含量。
中药材原料常含有较多的糖类成分,因此,在中药注射液生产过程中,特别是高温灭菌过程中,很容易产生己糖的降解产物5-羟甲基糠醛。而中药注射液直接进入体内循环系统,需严格保障其质量,以免发生不良反应。为了控制注射液的安全性,
